It will be easiest when they are side by side and the comparison is really only particularly useful for adults. The female will be slightly larger and more pear shaped. Of course, if you have a calling Ranitomeya, it's a male. The lack of a call doesn't guarantee female, though. There are images online for Ranitomeya in general.

This might sound crazy but I have had some success in the past with actually playing back territory calls from males with other similar species of dark frog and having the males reply if they are in a tank. I could try that as well.

Calling is really the only way, as with most Ranitomeya. Thankfully, R. vanzolinii do pretty well in groups.

If you have a ton of experience, you can probably make a pretty good guess at sexes based on size and behaviour, but calling is the sure fire way. Their call is similar to imitators, so very noticeable.
