How might I close a one inch gap between a top vent and the glass door? (short of getting another piece of glass cut...)

Are you describing an aquarium set up vertically? If so, a piece of acrylic can be attached to the vent from the back, but the latch will need to be redone to hold the glass door closed.
Do you have a picture? That would make it easier to make a recommendation.

Go to Hobby Lobby. Buy a piece of "quilting template plastic". Cut out what you need and attach with clear plastic packaging tape! Done!

I'm just getting new glass cut. It's a big pain but I would rather have it over with now rather than even bigger problems later...
